Le travail de la couleur est central dans toute œuvre artistique. C'est même l'un des premiers outils de l'artiste. Il est difficile d'imaginer une œuvre qui existerait sans le travail de la couleur - même si c'est l'absence de couleur que l'artiste choisit de présenter.
Au fil des époques et des mouvements artistiques, l'utilisation et le sens attribués à la couleur évoluent, mais l'essence de la couleur reste la même. Tout artiste se doit d'en maîtriser les propriétés pour pouvoir contrôler sa composition. Dans la restauration de tableaux, la couleur devient même une science, car il est nécessaire de connaître les différentes molécules pour retrouver les couleurs et mélanges utilisés à l'origine par l'artiste.
Dans l'histoire de l'art, l'importance de la couleur fluctue selon les périodes et les zones géographiques. Pendant la Renaissance italienne, par exemple, il existe un débat (appelé Paragone) entre l'autorité de la couleur par rapport à celle du dessin : selon les écoles, c'est la couleur, et non le tracé, qui crée l'émotion et le pouvoir visuel d'une œuvre d'art. Les couleurs prennent ainsi une importance immense, et endossent certaines significations : le blanc symbolise la pureté par exemple, et le bleu (systématiquement utilisés vêtir la Vierge Marie) est associé à la divinité. Ces symboliques ne sont pas pensées au hasard : le pourpre par exemple, est utilisé depuis l'époque Byzantine pour signifier le plus haut grade de royauté. Contrairement à l'ochre, le pigment pourpre provenait d'un coquillage spécifique, et était extrêmement difficile - et donc rare, et coûteux - à obtenir.
Plus généralement, les couleurs peuvent être réparties en trois catégories : les couleurs chaudes, les couleurs froides, et les neutres. Comme leur nom l'indique, ces classes de couleurs dégagent une atmosphère que le peintre peut utiliser pour influencer l'émotion de son œuvre. L'art baroque, par exemple, manipule les contrastes entre les couleurs chaudes et les couleurs froides pour saisir la puissance des corps. Les jeux de lumière sont exaltés par les effets de couleurs. Pendant longtemps, l'école occidentale traditionnelle de peinture exige que les peintres reproduisent les couleurs de l'environnement qui les entourent. Ce sont les impressionnistes, au 19ème siècle, qui explorent d'autres façon de voir - et donc de retranscrire sur la toile - leur environnement chromatique. En évitant les mélanges complexes et en peignant spontanément, en plein air, les impressionnistes réinventent l'utilisation de la couleur pour reproduire la réalité.
Il faudra atteindre la peinture abstraite et subjective pour que l'art se dévoue à la couleur comme sujet. Mark Rothko, précurseur du mouvement Colorfield Painting et de l'expressionnisme abstrait, voit dans ses tableaux un organisme vivant dont la couleur est humaine et le format transcendant. Piet Mondrian, lui, cherchera dans ses peintures à approcher l'essence même de la nature grâce à la pureté des couleurs primaires, pour atteindre l'abstraction. Le fondateur du mouvement d'avant-garde russe du Suprématisme, Kasimir Malévitch, troublera les sens de chacun avec son œuvre « Carré blanc sur fond blanc », dans laquelle la couleur n'est peinte que pour elle-même. L'art contemporain, la photographie, le collage, ou encore le pop art utilisent aussi à leurs manières respectives les ressources de la couleur, en explorant indéfiniment toutes ses pluralités. Comme disait Pablo Picasso, « Quand je n'ai pas de bleu, je mets du rouge ».
